The acting president would have been the president pro tempore of the Senate, Lafayette Sabine Foster of Connecticut.

The acting president would have been the President Pro Tempore of the Senate, Lafayette Sabine Foster of Connecticut. Senator Foster would have been acting president until March 4, 1866, and a special election would be held to fill the vacancy. If Foster wasn't available, Speaker of the House Schuyler Colfax would have been next in line. The rules have since changed and today the Speaker of the House supersedes the Senate President Pro Tempore.

If Andrew Johnson had been assassinated on the night of the Lincoln assassination in April 1865, we would have had President Lafayette Sabine Foster. Foster, a Senator from Connecticut, was president pro tempore of the Senate and next in line to the Presidency after Andrew Johnson.

Schuyler Colfax because he was the Speaker of the house at the time. https://www.history.com/news/10-things-you-may-not-know-about-the-lincoln-assassination

Ironically he was invited to Fords Theatre that night and declined. Grant and Robert Todd Lincoln also declined. As a sad note, Clara Harris and Major Henry Rathbone attended instead. Rathbone was stabbed by Booth and eventually he suffered deep psychological effects from that night. He was eventually admitted to an asylum.

Edit - Response revised in subsequent post.
